That photo taken by Phase One and professionally edited, but still I'm wondering if I can catch that much details with 5D III and 100mm L macro.

Focusing may always slow on some lenses, such as macro ones. What lens are you using?
Also can you try zone af and select the centre zone?

I have two IS lenses, 24-105 L and 100 L macro, I always turn off 24-105's IS if I don't need it, because it makes viewfinder jumpy when starting and stopping operation. I don't think you should worry about lifespan of it, as it's mentioned that it has a long lifespan, but the noise and jumpy viewfinder in some lenses.

Probably won't even cover a full frame sensor area. So not only are there little if any advantages for still photogs, there's a number of disadvantages.

Are you sure? Canon 1D C has a full frame sensor and produced for recording motion.

What kind of trip are you taking?  A 35 Liter pack isn't very big and it will be even smaller after you put in camera gear.  A 35 liter pack is usually only big enough for warm weather weekend trips.  Will you have to carry a tent, sleep gear, etc or are you just doing day hikes?  I really hope you are already an experienced hiker/camper so you know what you're doing on the trail.  Remember that back country pro photographers sometimes hire assistants to help them carry gear when necessary.  Don't over do the gear carrying it by yourself!

It's not serious camping, basically we are driving nearby the location we want to do camping, we are trekking around with backpacks but for sleeping we are going back to car to take tents and stuff. So it's not serious camping.

As you give pound prices I assume you live in the UK. You should be able to find it easily in here.
I got some of my Lee system on ebay, they were like new condition and was about £30 cheaper than new price.

I use manfrotto new 055 tripod, when it's fully extended without centre column it's taller than me.
So I do not need centre column, nevertheless I use it in indoor macros.

One of the main factors for me was F2.8 for night star landscape, as you can open the shutter longer and keep the stars sharp at 16mm, and its a zoom which I can also use for landscape and weddings. The 16-35mm doesn't do that… The F2.8 is very useful and if you haven't used it for that reason give it a go it will make you want to keep it, its very useful in a way the 16-35mm F4 isn't.

Excuse my lack of knowledge.
How could it make too much effect simply increasing ISO from 100 to 200, why not shoot with a f/4 lens in ISO 200?
